再生医療センター

再生医療センターについて

胚性幹(ES)細胞、人工多能性幹(iPS)細胞や体性幹細胞などの様々な細胞を用いた再生医療に関する研究を行っています。人のからだは事故や病気で機能を失うことがあります。臓器が一旦機能を失うと、その機能を回復させることは大変難しくなります。「再生医療」とは、他人の臓器そのものを移植するのではなく、細胞の移植を行うことにより、臓器の機能を補い、臓器を再生させることを目指した治療法です。再生医療について、幹細胞、ES細胞、iPS細胞を対象とし、その有効性、安全性を様々な観点から検証し、臨床応用の実現に向けた取り組みを行っています。


研究部紹介

細胞医療研究部

  • 受精メカニズムに関する研究を行っています。受精の膜融合過程は、精子の卵細胞膜への接着、融合、多精拒否からなる一連の現象です。本テーマではその分子メカニズムの解明に挑戦するため、膜融合過程に関わる因子群を明らかにし、その挙動を可視化することにより、受精における膜融合が時間的・空間的にどのように形成されるのか、解析しています。
  • ヒト組織幹細胞を生体外で培養し、増殖させることに成功しており、幹細胞の分離・同定を行っています。再生医療の材料となる細胞は、生体外における培養技術とそれによる細胞数の確保、それに続く分離技術が重要な要素となります。安全で質の高い細胞移植・再生医療の提供に向けた取り組みを行っています。
  • 生体肝移植は様々な場面で行われており、小児科領域においても一般的な医療としての地位を獲得しています。しかし、新生児や低体重児に対する生体肝移植については、課題が多いのが現状です。この現状を打破すべく注目されているのが、肝細胞移植です。本研究室では、肝細胞移植に特化した研究を行っています。これらの成果を基盤として国内初の肝細胞移植を実施しました。肝細胞移植の臨床応用に向けて着実に前進しています。

生殖医療研究部

  • ヒト幹細胞に関する研究を行っています。幹細胞には体性幹細胞、ES細胞、iPS細胞等様々な種類があり、それぞれ異なる特徴を有しています。京都大学に次ぐ国内2施設目のヒトES細胞樹立機関としての認定を受け、現在までに7株のヒトES細胞(SEES1-7)を樹立しました。また、病院部門と連携し、患者から同意を得た上で検体を提供していただき、その検体組織からの細胞の体性幹細胞の分離・培養技術を確立し、成育バイオリソースを構築しています。さらに、成育バイオリソースを利用したiPS細胞の作製、機能解析を実施しています。

研究内容

受精からヒトとして成長する過程で生じる疾患の成立機序の解明とその予防や、診断・治療法の開発を目指した研究を行っています。生殖の主体となる卵子、精子などの生殖細胞やES細胞・iPS細胞含む各種幹細胞を主な研究対象としています。さらに、生殖腺、胎盤、心臓、神経系、骨、軟骨、脂肪組織を研究対象に加え、幹細胞の機能を調節する分子機構の解明と臨床応用を目指した一連の研究を展開しています。これらの基盤的研究をさらに臨床研究に進展させることにより、生殖医療ならびに再生医療に貢献することが当研究部の使命と考えています。

「いのちの萌芽(受精)」のエビデンスに基づいた考え方の提示

受精の研究から、不妊治療への道が開かれるとともに、受精以外の膜融合にも新しい概念を提案できると考えています。

ヒト胚性幹細胞の樹立

「ヒトES細胞の樹立及び使用に関する指針」を遵守して、医学的、社会的にも適正に行なわれるよう、外部委員も含めたヒトES細胞研究倫理審査委員会(以下「倫理審査委員会」)を設置しています。現在、国の指針を遵守し、我々の研究がヒト生命の萌芽の滅失の上に成り立っていることを常に認識し厳粛にヒトES細胞研究を行っています。

細胞治療・再生医療

ヒト組織幹細胞を生体外で培養することで増殖させることに成功しており、その分離・同定を行っています。細胞の生体外における培養技術とそれによる細胞数の確保とそれに続く分離技術は、再現性の高い生物実験系に基づいた細胞の基盤解明によってのみ可能となるものです。

成育バイオリソースの構築

病院内の整形外科、産婦人科、眼科、形成外科と連携し、骨髄、臍帯血、胎盤、子宮内膜を細胞供給源として研究を行っています。臨床研究を前提としたad hoc委員会を設立し、臨床試験研究への具体的なロードマップの作成、医療に提供できる新たなヒト細胞の分離・培養法の開発、ヒト血清ならびにヒト液性因子のみからなる培養法の開発を行っています。
