研究部紹介

小児の難治性遺伝性疾患に関わる原因遺伝子を同定し、その機能を解析することで遺伝子ベースでの新たな治療法の開発を目指しています。具体的には、疾患iPS細胞の樹立とその解析ならびに臨床用ベクターの開発を行っており、これら研究結果の下に遺伝子治療臨床研究を準備しています。
特に原発性免疫不全症に対する、次世代シークエンサーを用いた遺伝子解析と疾患特異的iPS細胞を用いた病態解析による網羅的相補的遺伝子診断法(comprehensive complimentary diagnosis: CCD)の開発や新規治療法としての造血幹細胞遺伝子治療の実施は、当センター病院と一体となって進めている主要な研究テーマです。


研究内容

○原発性免疫不全症への遺伝子治療の確立

  1. X連鎖慢性肉芽腫症に対するレトロウイルスベクターによる遺伝子治療を米国国立衛生研究所(NIH)との共同研究にて行っています。
  2. Wiskott-Aldrich症候群やX連鎖重症複合免疫不全症に対してレンチウイルスベクターを用いた遺伝子治療の実施を準備しています。
  3. 遺伝子治療臨床研究の安全性を評価するため、臨床用ウイルスベクターの品質管理や遺伝子治療用ベクターの染色体挿入部位解析を次世代シークエンサーやdigital PCRを用いて行っています。

○疾患特異的iPS細胞の樹立

  1. 原発性免疫不全症や先天性代謝疾患などの小児難治性遺伝性疾患患者から採取した細胞(血液細胞や皮膚細胞)より、疾患特異的iPS細胞を樹立しています。
  2. 樹立したiPS細胞を用いて詳細な病態解析や遺伝子改変技術の研究を行っています。

○代謝疾患に対する遺伝子治療法の確立

Niemann-Pick病やムコ多糖症などのライソゾーム病を主な対象とし、アデノ随伴ウイルスベクターを用いた新規治療法の開発を行っています。

○ウイルスベクターに関する研究

  1. ヒト由来の転写抑制因子をクローニングし、レトロウイルスベクターの遺伝子発現抑制機構を解析しています。
  2. 将来の遺伝子治療臨床研究に向け、より安全で治療効果の高いウイルスベクターの開発を行っています。

○成育バイオバンク

小児難治性遺伝性疾患患者から得られたiPS細胞等を成育バイオバンクのバイオリソースとして保管し、創薬を含む今後の科学の発展に役立つバンク事業を目指しています。
