免疫アレルギー・感染研究部

研究部紹介

気管支喘息、アトピー性皮膚炎、食物アレルギー、川崎病といった様々な免疫アレルギー疾患や、移植時の拒絶反応、先天性サイトメガロウイルス感染症をはじめとした母児感染症や重症感染症が発生する仕組みについて、疫学研究・臨床研究・基礎研究の手法を駆使して解明し、これらの疾患に対する有効な予防・治療および診断法の開発を最終的な目標として研究を行っています。

 アレルギー疾患を発症する小児の割合は近年増加傾向にあります。このようなアレルギー疾患の増加には、主として環境因子と遺伝的素因が大きく関与していると考えられています。アレルギー研究室では、アレルギー疾患の発症や重症化の仕組みを明らかにし、それを応用した新たな発症予防法の開発を目指して研究を行っています。特に、近年新たに発見された自然リンパ球と組織構成細胞との相互作用に着目し、研究を行っています。また、近年報告が増えている好酸球性消化管疾患に関する病態解明研究も行なっています。

 川崎病は乳幼児に好発する原因不明の全身性血管炎です。特に、心臓に血液を供給する冠動脈の炎症による後遺症(冠動脈瘤の形成)を残すことが臨床上最も重要な問題となります。冠動脈障害が原因で心筋梗塞を起こすこともあり、小児の後天性心疾患の原因の一つです。また、未だ川崎病の病因や発症メカニズムが解明されていない中、日本の年間患者発生数は世界一多く、年々増加していることも疫学調査から明らかになっています。炎症制御研究室では、川崎病の急性期標準治療として用いられている免疫グロブリン療法(IVIG)の冠動脈炎症に対する抗炎症効果の作用機序について、分子細胞生物学的手法を用いた解明を目指しています。この研究成果は、IVIG治療抵抗例のような重症川崎病患者に対する新たな治療戦略の確立に役立つと考えています。

 移植免疫研究室では、様々な臓器移植モデル、やIn vitroの評価系を用いて、免疫寛容の誘導・維持に関与する分子・細胞の機序の解明およびCell Based Therapy を中心に、制御性T細胞、iPS細胞由来制御性樹状細胞(DC)、ミエロイド由来抑制細胞(MDSC)、間葉系幹細胞を用いる免疫制御細胞療法を確立するための基礎研究を行っています。また、新規免疫抑制剤、新規臓器保存法の開発を積極的に進めています。
母児感染研究室では、先天性サイトメガロウイルス感染症をはじめとした母児感染症について、新規感染モデルの確立、病原性発現機構の解明、および新規診断・治療法開発に向けた研究を行っています。

 感染症は、小児が罹患する疾患として最も頻度の高いものです。また、免疫不全症をはじめとする慢性疾患や手術後の患者における合併症としても感染症は重要です。感染免疫研究室ではリアルタイムPCRなどを用いた感染症の診断法の開発および実際の診断を行っています。研究成果は診療への直接の還元のみならず、病態解明や新しい治療法の評価指標として活かされています。
